Shinzaburo Noguchi is a co-author of the two BOLERO-2 papers in the Atlas. BOLERO-2 was a phase 3 randomised trial in 724 postmenopausal women with hormone-receptor-positive, HER2-negative advanced breast cancer that had progressed on an aromatase inhibitor. Resistance to hormone therapy in these tumours is linked to overactive mTOR signalling. All women received the hormone drug exemestane, and half also took everolimus, an mTORC1 inhibitor related to rapamycin.
BAS2012, in the New England Journal of Medicine, showed that everolimus more than doubled the time before the cancer grew again, from 4.1 to 10.6 months by central review, which led to approval of the combination. Mouth sores (stomatitis) were the main side effect. PIC2014, in Annals of Oncology, gave the final overall survival: 31.0 months with everolimus and 26.6 months with placebo, a difference that was not statistically significant. The trial showed that blocking mTOR can slow a common cancer, but did not prove that it extends life.
PubMed lists well over a hundred breast cancer papers under this name from the Department of Breast and Endocrine Surgery at Osaka University, up to 2023. No official profile was opened, so the current title is not given here.
