BOLERO-2 was a phase 3 trial in 724 postmenopausal women with hormone-receptor-positive advanced breast cancer whose disease had grown despite an aromatase inhibitor, a standard hormone therapy. Resistance to hormone therapy is linked to mTOR activation, so the trial added everolimus, an mTOR-blocking drug related to rapamycin, to the hormone drug exemestane. In the first report (2012) the time before the cancer grew again, called progression-free survival, rose from 4.1 to 10.6 months by central review, and the combination was approved by the FDA. The final 2014 analysis found that overall survival was longer on paper (31.0 versus 26.6 months) but the difference was not statistically significant. This is direct evidence in people, for one type of advanced cancer.

Alejandra Perez is one of the trial's investigators, the doctors who enrolled and treated patients, and is a co-author of both reports. Perez also co-authored follow-up BOLERO-2 analyses, including a 2014 paper on when everolimus side effects appear and a 2015 study of which tumour gene changes went with benefit from everolimus.

Perez trained in medicine in Medellin, Colombia (1991), did a residency in internal medicine at Danbury Hospital, Connecticut, and a fellowship in hematology and oncology at Montefiore Medical Center in New York (2000). For many years, Perez worked at the Breast Cancer Centers of the Memorial Cancer Institute in Hollywood, Florida, where Perez was first author of a 2010 early-phase trial of BZL101, an extract of the herb Scutellaria barbata, in metastatic breast cancer. Perez now treats breast cancer at the University of Miami's Sylvester Comprehensive Cancer Center in Plantation and served on the panel for the 2021 ASCO guideline on treatment before breast cancer surgery.
