Lysosomal cholesterol activates mTORC1 via an SLC38A9-Niemann-Pick C1 signaling complex
What this study shows
Establishes lysosomal cholesterol as a nutrient input that drives mTORC1 recruitment and activation at the lysosomal surface. SLC38A9 is required for cholesterol-driven mTORC1 activation through conserved cholesterol-responsive motifs, and does so independently of its arginine-sensing function; NPC1, which exports cholesterol from the lysosome, binds SLC38A9 and restrains mTORC1 through its sterol transport function. Boundary: cell lines and patient-derived fibroblasts, no organismal endpoint.
At a glance
| Evidence type | M Molecular — cells, biochemistry, structure Marked M because it is molecular or in-vitro work (model: Human cell lines (HEK293T), CHO cells, NPC patient fibroblasts) rather than a whole-organism health-outcome study. That is often exactly where causal biology gets established -- the code says which system the finding was shown in, and nothing about how good the work is. |
