Matthew Sherman was senior author, on the sponsor side at Wyeth, of the 2004 randomised phase II trial of temsirolimus (CCI-779) in advanced, previously treated kidney cancer (ATK2004). Patients received 25, 75 or 250 mg weekly; the response rate was 7% and neither efficacy nor toxicity differed clearly by dose, which supported the 25 mg dose used later. The trial had no control arm.

Sherman trained in chemistry at MIT, in medicine at Dartmouth, in internal medicine at Georgetown and in medical oncology at Dana-Farber, and held academic posts at Harvard Medical School. He spent more than ten years at Wyeth-Ayerst Research and Genetics Institute, where he also worked on the antibody-drug conjugate gemtuzumab ozogamicin for acute myeloid leukaemia, and later served as chief medical officer at Synta Pharmaceuticals, Acceleron Pharma and Deciphera Pharmaceuticals.
