Ke Hu is the first author of HU2026B, from Stefan Feske's lab at NYU Langone, identifying the sodium-bicarbonate cotransporter SLC4A7/NBCn1 as a selective regulator of antibody-producing plasmablast differentiation. The study showed that SLC4A7 keeps the inside of a differentiating B cell from becoming too acidic; without it, cells acidify internally, their lysosomes become abnormally alkaline, and mTORC1 activity falls, blunting antibody responses in mice after immunization and influenza infection. Restoring mTORC1 activity, or acidifying the cells by blocking a different transporter (the Na+/H+ exchanger), rescued differentiation, tying this pH-sensing step directly to mTORC1 function in humoral immunity.
