Elena Belousova Portrait: Neuro-Med clinic (nevromed.ru)

Tuberous sclerosis complex (TSC) is a genetic disease in which a faulty TSC1 or TSC2 gene leaves mTOR permanently switched on. Children and adults with TSC grow benign tumours in many organs, including the brain and the kidneys, and most have epilepsy. Elena Belousova is a co-author of both large phase 3 trials of everolimus, a drug that blocks mTORC1, in this disease. In EXIST-1 (FRA2013), 117 patients with brain tumours called subependymal giant cell astrocytomas took everolimus or placebo; the tumours shrank by at least half in 35 percent of patients on the drug and in none on placebo. In EXIST-2 (BIS2013), 118 patients with kidney tumours called angiomyolipomas took part; 42 percent on everolimus had a response, against none on placebo.

These were randomised, placebo-controlled trials in people, and they are among the clearest evidence that blocking mTOR works when a genetic fault is the cause of mTOR overactivity. That does not mean the same benefit applies to people without such a mutation.

Belousova trained in medicine at the Leningrad Pediatric Medical Institute, graduating in 1980, and completed a residency in child neurology in Leningrad. She has worked since 1985 at the Moscow Research Institute of Pediatrics and Pediatric Surgery and has headed its Department of Psychoneurology and Epileptology since 2003. Her doctoral thesis (2004) was on seizures and epilepsy in children with cerebral palsy, and she became a professor in 2007. Her work covers the diagnosis and treatment of childhood epilepsy, and she has been a principal investigator in international trials in tuberous sclerosis and in Duchenne muscular dystrophy. She was named Honored Physician of the Russian Federation in 2011.
