Dianqing Wu is a pharmacologist at Yale. His research looks at how cells pass on signals from receptors at the cell surface, especially the Wnt pathway, which matters in development, bone and cancer, and the signals that guide white blood cells to move toward sites of inflammation.
In the 2011 study in this Atlas, with first author Xiaoqing Gan, the team asked whether the lipid PIP3 only acts upstream of mTORC2 or can switch it on directly. In cells, insulin and active PI3K raised phosphorylation of the Akt hydrophobic-motif site, and a PI3K inhibitor blocked it. In test-tube assays, PIP3 stimulated mTORC2 complexes to phosphorylate Akt.
The result links PI3K signalling to mTORC2, one of the two main mTOR complexes. It was done in cultured cells and in vitro.
