Delong Meng

Cancer researcher at UCSF who led the preclinical test of RMC-6272, a drug that blocks mTORC1 much more strongly than mTORC2, in drug-resistant breast cancer

Department of Bioengineering and Therapeutic Sciences, University of California San Francisco (at the time of the 2023 study, with Sourav Bandyopadhyay)

Delong Meng is the first author of MEN2023, an Oncogene paper from Sourav Bandyopadhyay's group at UCSF, written with colleagues at Revolution Medicines, the company that makes the drug. mTOR works in two complexes. mTORC1 drives growth, and in many breast cancers it stays switched on because of mutations in PIK3CA or PTEN. Older drugs either block mTORC1 only partly, like rapamycin, which misses the target 4E-BP1, or block both complexes, which causes side effects such as high blood sugar. RMC-6272 is a bi-steric inhibitor: one molecule that grips mTOR at two sites at once. It is more than 25 times more selective for mTORC1 than for mTORC2 and shuts mTORC1 down completely, including its control of 4E-BP1.

In the study the drug killed ER-positive breast cancer cell lines, especially those with PIK3CA or PTEN mutations, and still worked in models that had become resistant to hormone therapy and to CDK4/6 inhibitors. The tests used cell lines and patient tumours grown in mice (PDX models), so they are preclinical. The paper laid groundwork for the related drug RMC-5552, which has gone into clinical trials.

PubMed shows no other paper that can be tied to this name with the same co-authors, and no official profile was found, so the current position could not be confirmed. The affiliation shown is the one on the Atlas paper.
