Boglarka Rauch
Researcher in Péter Bai's group at the University of Debrecen who studies how ADP-ribosylating enzymes control fat cell development through AMPK and mTOR
Department of Medical Chemistry, Faculty of Medicine, University of Debrecen (papers 2019–2026)
Fat cells form from precursor cells in a process called adipocyte differentiation, which needs mTORC1, the growth-control complex, to be active. In RAUCH2026, Boglarka Rauch and colleagues in Péter Bai's group worked on tankyrases, enzymes that attach chains of ADP-ribose to other proteins and so change how those proteins behave. In a cell line, deleting tankyrase-1 or tankyrase-2 slowed fat cell formation. The cells showed active AMPK, the energy sensor, low mTORC1 activity and blocked autophagy. The defect caused by losing tankyrase-2 could be fixed by NV-5138, a drug that switches mTORC1 on, or by blocking LKB1, the kinase that activates AMPK. Tankyrase-2 was found to ADP-ribosylate LKB1, which suggests that it normally keeps the LKB1-AMPK brake on mTORC1 in check. The findings come from cultured cells only.
Rauch has published with the Bai group in the Department of Medical Chemistry at the University of Debrecen since 2019, including a 2023 screen of bacterial metabolites for effects on breast cancer cell growth. | 2026 | M | Tankyrase-2 regulates adipocyte differentiation through AMPK/mTOR signaling. RAUCH2026 First author: showed in a cell line that tankyrase-2 supports fat cell differentiation through the LKB1-AMPK-mTORC1 axis and ADP-ribosylates LKB1. |
