Pathway link TSC-RHEB

TSC1/TSC2 Rheb

TSC1/TSC2 inhibits Rheb.

TSC2 forces Rheb to switch itself off – the one step where TSC acts as a tumour-suppressing brake.

TSC2 is a GAP for Rheb: it forces Rheb to hydrolyse its GTP, switching it off. This is the single step where the tumour-suppressor function lives.

The conditions it holds under

In vitro, Drosophila and mammalian cell biochemistry (INOK2003, GAR2003). GAP activity is shown in cells and in vitro; not measured in human tissue by these sources.

Step
direct (one molecule acts on the other)
Measured in

The source works as a GAP: it changes the GTP or GDP state of the target. The label says what that does to the target, not that the target is switched on or off everywhere.

“How settled” describes how far researchers in the field agree. It is not the same as the sign-off above, which only says whether a named curator has checked this page.
