Samuel Peña-Llopis
Cancer geneticist who, in James Brugarolas's lab, found that mTORC1 controls expression of lysosomal V-ATPase genes through TFEB; his group now studies BAP1-mutant uveal melanoma, kidney cancer and cholangiocarcinoma.
Group Leader, Translational Genomics, Department of Ophthalmology, University Hospital Essen, and member, Center for Medical Biotechnology (ZMB), University of Duisburg-Essen · DKTK partner site Essen · previously Brugarolas lab, UT Southwestern Medical Center, Dallas
Samuel Peña-Llopis was first author of the 2011 EMBO Journal study from James Brugarolas's lab at UT Southwestern (PEN2011). Comparing Tsc2-null and wild-type mouse cells with and without rapamycin, the team found genes that follow mTORC1 activity, enriched for subunits of the lysosomal V-ATPase, and showed that a large share of them needed TFEB. In these low-serum conditions active mTORC1 increased nuclear TFEB, the opposite direction to later studies in fed or starved cells.
Peña-Llopis now leads the Translational Genomics group at University Hospital Essen. His group works on cancers with BAP1 mutations, including uveal melanoma, renal cell carcinoma and cholangiocarcinoma, and uses patient-derived organoids to test drugs.
