Luis F. Parada Portrait: Memorial Sloan Kettering Cancer Center

Luis Parada is a neuroscientist and cancer biologist at Memorial Sloan Kettering Cancer Center, where he directs the Brain Tumor Center. His lab builds mouse models of nervous system tumours, including glioblastoma, neurofibromas and malignant peripheral nerve sheath tumours. When the Atlas study was done in 2009 his lab was at UT Southwestern Medical Center in Dallas.

The 2009 paper in the Atlas, with Jing Zhou as first author, used mice that lack the PTEN gene in some neurons of the cortex and hippocampus. PTEN normally holds back the PI3K and AKT pathway upstream of mTOR, so these mice have overactive mTORC1, enlarged neurons, large brains and behaviours that resemble some features of autism.

Rapamycin, an mTORC1 inhibitor, both prevented and reversed the neuron enlargement and improved some of the abnormal behaviours. The work pointed to mTORC1 as a drug target for PTEN-related brain disorders. It was done in mice.
