Ronald S. Duman

Psychiatric neuroscientist who showed that ketamine's rapid antidepressant effect depends on mTOR signaling and the growth of new synapses in the prefrontal cortex.

Deceased. 1954–2020 · Professor of Psychiatry and of Neuroscience, Yale School of Medicine · Yale Psychiatry from 1988

Ronald Duman (1954 to 2020) was a neuroscientist at Yale School of Medicine, where he joined the Department of Psychiatry in 1988. With Eric Nestler he co-founded the Laboratory of Molecular Psychiatry. He died suddenly in February 2020 while hiking near his home in Connecticut.

He proposed that stress weakens the growth-factor signals that keep brain cells and their connections healthy, and that antidepressants work in part by restoring them. He also showed that antidepressants promote the birth of new neurons in the adult hippocampus.

The 2010 Science paper in the Atlas, led by Nanxin Li, found that ketamine rapidly activates mTOR in rat prefrontal cortex and increases the number of synapses. Blocking mTOR with rapamycin stopped both the new synapses and the antidepressant-like behavior.
