Jacqueline A. French
Epilepsy clinical-trial specialist; first author of EXIST-3, the placebo-controlled phase 3 trial in which everolimus reduced treatment-resistant seizures in tuberous sclerosis complex.

Jacqueline French was first author of the EXIST-3 trial (FRE2016). 366 patients aged 2 to 65 with tuberous sclerosis complex and focal seizures that had not responded to standard drugs were randomised to placebo or to everolimus at a low or high blood level, added to their usual antiepileptic drugs. At least a 50% fall in seizure frequency was seen in 15.1% on placebo, 28.2% on low exposure and 40.0% on high exposure, with more adverse events at higher exposure. The trial showed a dose-dependent effect of mTOR inhibition on seizures in TSC over an 18-week core phase. It builds on mouse work such as ZEN2008.
French trained at Brown, Mount Sinai and Yale, was a professor of neurology at the University of Pennsylvania, and moved to NYU Langone in 2007. Her work centres on how antiseizure drugs are tested: she founded the Epilepsy Study Consortium, an academic network for early-phase epilepsy trials, and helped develop trial designs accepted by the FDA.
